000585114 520__ $$aStructural biology offers a window into the functionality of molecularmachines in health and disease. A fundamental challenge lies in capturingboth the high-resolution structural details and dynamic changes that areessential for function. The high-resolution methods of X-ray crystallographyand electron cryo-microscopy (cryo-EM) are mainly used to studyensembles of static conformations but can also capture crucial dynamictransition states. Here, we review the latest strategies and advancementsin time-resolved structural biology with a focus on capturing dynamicchanges. We describe recent technology developments for time-resolvedsample preparation and delivery in the cryo-EM and X-ray fields and explorehow these technologies could mutually benefit each other to advance ourunderstanding of biology at the molecular and atomic scales.
